Yes...you can...
Click Start, then Run, type cmd and click "Ok".
At the command prompt, type convert X: /fs:ntfs (where X is the letter of the drive you wish to convert...note the spaces in the command) and press "Enter".
This procedure should not bother your data, but I always recommend current backups before performing an operation of this kind.
