What does this mean and what can I do about it? I always press "no" because I do not know what it is for and it still comes up every time I start up firefox.

You have the Options set to have FF check if it is the default browser you use. If you use FF all the time, then click yes the next time you get the Alert box and you'll not see it again. If you do not use FF that often, then go into Tools/Options/Main tab and under System Defaults, uncheck box to have FF check if it is default browser on startup.
