Serious virus infection

I am working on a laptop with a serious virus infection. I have ran GMER, Root Repeal, Malwarebytes, Symantec Vundo Remover, and Smitfraudfix on the computer and it still is infected. Malwarebytes scans show infected items and says it removes them but it does not. I am at least able to access a desktop now after running Smitfraudfix on the machine. Before that I could only run apps from the SafeMode command prompt. I need help!
