Why is a system reboot required after installing or uninstalling something and also exactly when a system reboot is required?

Some changes to the operating system can only be done on shutdown and/or startup.


... and also exactly when a system reboot is required?

Whenever a change to the operating system would require a reboot in order for that change to take effect.

Does that also apply to any changes to the system settings?

Not necessarily to system settings. I've had changes to the page file ask me to reboot before (and sometimes not - it's weird). Usually when a program uninstall needs a reboot, it's not because of a shared component or something like that, but because the program uses it's own installer and can't delete itself without telling the operating system to delete the file on startup. Something along those lines.
