Yes I'm happy that I've recovered all of the data I need from the hard drive so a clean wipe wouldn't be a problem

Please confirm whether or not you still get the same message on the computer that has issues.

Can you also answer the above for us.

What I would do first is check that the boot sequence is set to DVD/CD drive first and then see if Puppy Linux can still access the data on the drive, if it can it makes it less likely (but not impossible) that it is the DVD drive that is not reading the ISO, Puppy will run on a CD only drive whereas Windows 10 needs a functional DVD drive due to the video content it contains, in short, the CD part of an optical drive can be working perfectly but quite often the laser on the DVD part of the drive fails and DVD content cannot be read or burned.

Does Edit Boot Options screen appears when you press F10 function key if it does use VAIO Hardware Diagnostics utility.
Use Qemu Simple Boot tool to test if Windows 10 USB flash drive or DVD is bootable.
Did you press the Assist button while the power was off?

F10 doesn't seem to do anything on startup, neither tapping nor holding down.

The only thing that works is holding the assist button to power on. It then goes to a white vaio care (rescue mode) screen with various bios and startup options

Just tried the DVD on the older working laptop and received the "press any key to boot from disk" message

Please confirm whether or not you still get the same message on the computer that has issues.

You keep missing answering this question for some reason mackemmatt.

No problem and thanks for confirming

Not seen the QEMU Simple Boot software suggested by FreeBooter but it does look worth giving it a go at checking your ISO out.

The only thing that works is holding the assist button to power on. It then goes to a white vaio care (rescue mode) screen with various bios and startup options

Good this is where you can boot from DVD or USB flash drive or factory reset your computer or try repair your computer with Windows recovery tools like system restore tool or Startup Repair tool.

With QEMU Simple Boot utility USB flash drive can be virtually tested to see if USB flash drive is bootable or test CD/DVD media or ISO image.

Not sure if i've done this correctly but I downloaded QEMU simple boot, added the ISO from my hard drive and ran the test to receive the pop up below. Do I need to adust the ram size or have I not created the ISO correctly?

Increase the RAM size if you have more spare RAM left running on your system.

Increasing the ram size to 512mb gave this screen

qemu 512 mb.png

Does this suggest the ISO is functioning correctly?

Yes it does for some reason QEMU simple boot have trouble booting Windows 10 but that screen proves Window 10 bootable.
