Cannot access anything

Yesterday something took complete control of my computer. I cannot access anything on my computer. When I try I get random exe files infected messages, do you want to fix now. The only thing that will open on the computer is a screen looks like IE on THEIR software page for purchase. I have malwarebytes installed as well as McAfee so this has disabled all my security settings. I of course am not being allowed to open them. I did open in safe mode and ran them both with no infection detected. I need to know what and how I can access any information you need to help me. Running Vista.
