Blocking Attempts to Add/Alter Registry Values

Lavasoft's Ad-Aware has an Ad-watch alarm listing all attempts to add registry values. I don't often know which ones to block.
Should I block First Home Page (Software\Microsoft\Internet Explorer\Main)?
It looks like an attempt to force a certain home page upon Explorer start-up?
In that case, I want to block it.
However, I am a novice user.
What do others suggest?
Where can I find a guide on what to block? the Lavasoft web page does not get specific.
