USB connections aren't working.

So I just built my new computer and I don't think I connect things right, but yet I do because when I connect my monitor to the motherboard it shows up like somethings connected to it. When I put anything USB related like my mouse, keyboard, etc. it will light it for a mere second or two and then go away. But I have a question to as weather or not I have to have my OS installed already. My DVD drive is shot because I used one from my ollllllld computer so I couldn't put the CD in and I was told I could install it using a flashdrive. But my flashdrive doesn't work. So any input on this would help tremondously.
