As long as the video card is PCI Express, and you have the power supply it should run no problem.

Well, what I meant to ask was, are there two different variants of video cards out now that PCI express is available? As in, an AGP variant and a PCI variant? Or will the card designed for the AGP slot work in the PCI express slot?

Ok, there are two types of PCI Express slots. PCIx16, and PCIx8, the x16 is for video, and the x8 is for addin cards(Sound, NIC, and so on). You cant place a AGP card in any PICx slot, but you can use the old PCI cards in the PCIx8, but the PCIx8 cards cant go in to the old PCI slots:
AGP -> PCIx16 = No
PCIx16 -> AGP = No
Old PCI -> PCIx8 = Yes
PCIx8 -> Old PCI = No
