You need to boot into recovery console with your windows 2000 CD and then make the change there.
Warning: You did not say why you want to do the above, and since you do not know what the recovery console is, I suspect your PC knowledge is somewhat limited and I fear that you are about to really mess up your machine.

i want to change user32.dll i have edited the file now i want t replace it ,if i boot from a win cd then also it wont go in dos mode idf there is any way plz tell me in detail thankyou

once again, I urge you to stop, think, reconsier. That is an ultra critical windows file. You will KILL your computer if you screwed up the edit that probably was a bad idea in the first place.

if you know what you're doing, how is it you do not know how to load the recovery console. This is pretty basic stuff.
A lot of people think they know what they are doing and screw their computers up hopelessly.
